Critical Airflow Visualization (CAV) & Smoke Study Services

CAV studies are a formal regulatory requirement for pharmaceutical and biotech manufacturers operating aseptic environments under FDA and EU GMP Annex 1, and the standard goes beyond confirming a facility is well designed.  CAV’s Role in Aseptic Manufacturing

Critical airflow visualization, commonly referred to as a smoke study, uses smoke or fog to characterize and document airflow patterns in aseptic manufacturing environments. Studies confirm that airflow in critical processing areas moves in the intended direction and protects open product and container closure systems from contamination, producing the objective evidence regulators require that a facility maintains the proper cleanroom environment.

CAV studies are required before initiating manufacturing in a new or modified facility, following significant changes to equipment or HVAC systems, and as part of ongoing sterile manufacturing qualification programs.

Sterility Assurance Requires More Than Static Testing

A well-designed facility can complete static testing without difficulty. What regulators require goes further: evidence that the facility, its equipment, and its operators function together to maintain required aseptic conditions under dynamic conditions, using the facility exactly as it operates during production. 

Static testing alone does not demonstrate that, and uncharacterized airflow creates contamination pathways that put sterility assurance, product quality, and compliance at risk.

Step 1: Project Planning

Every CAV engagement begins with a review of the facility layout, manufacturing operations, equipment design, and any prior study history. PV uses this assessment to develop a study plan covering all critical zones, intervention types, and dynamic activities required.

Step 2: Protocol Development

PV develops a written protocol documenting study scope, areas to be tested, intervention activities, acceptance criteria, and documentation requirements, structured to satisfy FDA, Annex 1, and applicable regulatory expectations.

Step 3: Pre-Study Assessment

Before testing begins, PV visually assesses the facility and equipment for potential airflow risks. Identifying and addressing problems at this stage avoids discoveries during qualification and allows PV to work effectively within tight availability windows.

Step 4: On-Site Execution & Smoke Generation

PV deploys its multi-camera recording system and CRF4 fogger to execute the study per the approved protocol. Dynamic activities are performed as they occur during actual manufacturing operations, with live streaming allowing remote stakeholders to participate in real time.

Step 5: Data Analysis & Review

Recorded footage is analyzed to characterize airflow patterns, evaluate each intervention, and identify areas requiring remediation. PV reviews and edits footage on-site without file transfer, preserving data integrity from capture through final documentation.

PV delivers a structured summary report documenting study design, each intervention evaluated, airflow characterization findings, and remediation recommendations where applicable, with full supporting footage and documentation written for regulatory review and inspection readiness.

Critical Airflow Visualization FAQs

Critical airflow visualization, commonly referred to as a smoke study, uses glycol-based fog and clean water or water for injection (WFI) fog to characterize and document airflow patterns in aseptic manufacturing environments. It demonstrates that airflow in critical processing areas, including cleanrooms, isolators, and Grade A zones, moves in the intended direction and protects open product and container closure systems from contamination.

CAV studies are required before initiating sterile manufacturing in a new or modified aseptic environment, following significant changes to the facility, equipment, or HVAC systems, and as part of ongoing qualification programs. FDA and EU GMP Annex 1 both require documented airflow characterization under dynamic conditions with written conclusions for each intervention evaluated.

Static testing characterizes airflow in an unoccupied facility with equipment in place but not operating. Dynamic testing characterizes airflow under actual manufacturing conditions, with personnel present, equipment in operation, and all interventions performed as they occur during production. Regulators require dynamic testing because static results alone do not demonstrate that aseptic conditions are maintained during real manufacturing operations.

CAV study requirements appear in FDA guidance for aseptic processing, EU GMP Annex 1 for the manufacture of sterile medicinal products, and USP general chapters applicable to sterile manufacturing. Each framework requires airflow visualization under dynamic conditions with documented written conclusions.

Any aseptic manufacturing environment where open product or container closure systems are present requires airflow characterization. This includes filling lines, lyophilization loading and unloading areas, locally controlled environments, stopper-processing equipment, and all Grade A-classified areas.

A complete CAV study produces a structured summary report documenting study design, each intervention evaluated, airflow characterization findings, and remediation recommendations where applicable. Digital recordings are included, with all documentation structured for regulatory review and inspection readiness.
